چکیده

KAREN CARPENTER, RODNEY h4IDDElDN multifunctional enzyme Pure LCHAD. with specific activity 30.4pmol 3-ketohexacatalysing three steps of decanoyl-CoA/min/mg, also showed ECH and KAT activities of 92.9pmol 2-octenoyl-CoA/min/mg and 4 . 3 ~ 0 1 J-ketodecanoylCoA/min/mg respectively. All 3 enzymes were active with acyl-CoA J. POLLI’lT* and BRUCE substrates of chain length from Q to c16 but no significant activity was detectable with substrates. Therefore “LCHAD” appears to be a multifunctional B-oxidation enzyme of lonn-chain specificity and Department of Biochemistry. Expression of novel isoforms of carnitine palmitoyltransferase I (CPT-1) generated by alternative splicing of the CPT-ibeta gene.

Carnitine palmitoyltransferase I (CPT-I) catalyses the rate-determining step in mitochondrial fatty acid beta-oxidation. The enzyme has two cognate structural genes that are preferentially expressed in liver (alpha) or fat and muscle (beta). We hypothesized the existence of additional isoforms in heart to account for unique kinetic characteristics of enzyme activity in this tissue. Hybridizatio...

Impact of the intramitochondrial enzyme organization on fatty acid oxidation.

The enzymes of mitochondrial beta-oxidation are thought to be organized in at least two functional complexes, a membrane-bound, long-chain-specific beta-oxidation system and a matrix system consisting of soluble enzymes with preferences for medium-chain and short-chain substrates. This hypothesis is supported by the observation that the inactivation of long-chain 3-ketoacyl-CoA thiolase by 4-br...

Peroxisomal beta-oxidation of branched chain fatty acids in rat liver. Evidence that carnitine palmitoyltransferase I prevents transport of branched chain fatty acids into mitochondria.

Fatty acid beta-oxidation was investigated in highly purified mitochondrial and peroxisomal preparations from rat liver. Under isotonic conditions, pristanic and homophytanic acid beta-oxidation in purified peroxisomes was severalfold greater compared to the oxidation in purified mitochondria. Branched chain fatty acid beta-oxidation in purified mitochondria was very low, and the oxidation was ...

On the Mechanism of Beta-oxidation of Long Chain Fatty Acids by Liver Mitochondria from Normal and Alloxan-diabetic Rats.

An accepted mechanism for mitochondrial oxidation of fatty acids, i.e. ,8 oxidation, although inferred to apply also to long chain fatty acids (>C&), has actually been elucidated only for relatively short chain acids (<C’S); only the activation (acyl coenzyme A synthetase) and initial dehydrogenation (acyl coenzyme A dehydrogenase) steps have been demonstrated for palmitate (1). The present pap...

Role of long-chain fatty acids in the postnatal induction of genes coding for liver mitochondrial beta-oxidative enzymes.

Introduction The regulation of specific gene expression in response to changes in nutrition has become a major aspect of modern nutrition, due to the development of molecular biological techniques that have allowed the cloning of a number of genes involved in the regulation of carbohydrate and fat metabolism. It has been shown that major (glucose, fatty acids, amino acids) or minor (iron, vitam...
